Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. This MAb selectively interferes with lymphocyte binding to lymph node, mucosal and synovial endothelium. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. CD28 is also expressed on human fetal NK cells and some NK cell lines, whereas on murine NK cells the CD28 expression is much broader.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CD11b is a cell adhesion molecule that acts as a receptor for cell surface ligands such as intracellular adhesion molecules (ICAMs) or soluble ligands. Integrins are heterodimeric proteins that contain an alpha chain and beta chain. IntegrinαM combines with the Integrinβ2 to form a leukocyte-specific integrin referred to as macrophage receptor 1 (Mac-1), or inactivated-C3b (iC3b) receptor 3 (CR3). IntegrinαM/β2 is important in the adherence of neutrophils and monocytes to stimulated endothelium, and also in the phagocytosis of complement coated particles. The protein CD11b has been implicated in the various adhesion-related interactions of cells such as monocytes, macrophages, natural killer (NK) cells, and granulocytes. It is part of a heterodimer that consists of CD11b and CD18. It also modulates the uptake of complement-coated particles within the cell. CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular scavenger receptor cysteine-rich (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. In Western blotting, this antibody reacts with two bands of ~MW of 210 kDa and 300 kDa, identified as two isoforms of Tenascin C. Specificity of this MAb is validated by sequential immunoprecipitation with a PAb against Tenascin C. Tenascin C is a multifunctional, disulfide-linkedhexameric extracellular matrix glycoprotein expressed in association with mesenchymal epithelial interactions during development and in the neo-vasculature and stroma of undifferentiated tumors.
